To study the functional consequences of RASSF1A expression in human cells, we made use of a tetracycline-inducible expression system (32) and the A549 lung cancer cell line, which is deficient in endogenous RASSF1A, p14Arf, and p16Ink4a expression (Supplementary Fig. S1A; ref. 35). In this model, addition of doxycycline induced transgenic expression of RASSF1A, which inhibited growth of lung cancer cells in vitro (Fig. 1A) and in vivo after s.c. transplantation into immunodeficient NOD/SCID mice (Fig. 1B).
