Intracellular calcium signaling is regulated by PLC?, which generates inositol 1,4,5-trisphosphate and diacylglycerol from PIP2. Inositol 1,4,5-trisphosphate stimulates the release of intracellular calcium stores upon binding to its receptor in the ER. The depletion of ER calcium stores then triggers extracellular calcium influx. Diacylglycerol and intracellular calcium signals cooperate to activate PKCs, which then activate other pathways such as the NF-?B pathway, ultimately leading to mast cell degranulation and cytokine production.
