When P6 cerebellar glia transfected with plasmids encoding the erbB2-Luciferase reporter, measured level of luciferase activity in glia alone or in glia after the addition of neurons indicated that neuronal contact induced a 2-fold increase in erbB2 promoter activity. The erbB2 promoter activity was completely abolished by cotranfection with dominant-negative Notch1 (OCDN1). Constitutively active Notch1 (FCDN1) expression induced a 1.75 +/- 0.23-fold ( p < 0.007) increase of erbB2 protein in glia. These results suggest that Notch1 upregulated erbB2 expression.
