  sub:_4 prov:value "To examine how BATF-mediated negative control of AP-1 activity influences specific transitions in thymic T cell development, the p56lck proximal promoter was used to drive expression of an HA Ag-tagged BATF protein in mice. Two founder animals positive for the transgene were bred to generate the FVB-lck line 2 and FVB-lck line 4 mice used in these studies (Fig. 1A).  To determine whether the overexpression of BATF influences cytokine gene expression, thymocytes and splenocytes from wild-type and transgenic FVB-lck line 2 littermates were stimulated in vitro with plate-bound anti-CD3{epsilon} mAb. Total RNA was isolated after 24 h, and RPA was performed. The induction of IL-2, IL-4, IL-5, IL-10, and IL-13 mRNA by transgenic thymocytes is impaired relative to that in wild-type controls, while the expression of IL-6 and IFN-{gamma} mRNA is comparable (Fig. 2A). Transgenic splenocytes showed reduced expression of IL-4 and IL-5 and enhanced expression of IL-2 (Fig. 2A). The RPA was repeated with RNA prepared from FVB-lck line 4 animals, and similar results were obtained (data not shown)." ;
