Fig. 5 A shows that mitochondria isolated from the livers ofSod2 ?/+ mice undergo more rapid swelling in the presence of calcium and t-butylhydroperoxide based upon the decrease in relative absorbance. To ensure that the increased rate of mitochondrial swelling was due to the induction of the permeability transition, cyclosporin A, an inhibitor of the permeability transition pore (42), was added to the reactions. As illustrated in Fig. 5, the addition of cyclosporin A inhibited the decrease in absorbance, indicating that the changes in absorbance were due to the induction of the permeability transition pore.
