We then analyzed the effect of BAF57 depletion on the expression of endogenous ER target genes, monitoring mRNA levels by quantitative reverse transcription-PCR (QRT-PCR). The mRNA expression levels of a number of well characterized ER target genes (pS2, cathepsin D, progesterone receptor, BRCA1, BRCA2, and bcl-2) increased in ZR-75-1 cells in response to treatment with 17beta-estradiol (E2) for 24 h, as shown in Fig. 1C. The E2-dependent activation of all of these genes was markedly impaired by transfection with siBAF57 (Fig. 1C), indicating that BAF57 is important for the transcriptional activity of endogenous ER.
